Manatsu Natsuumi moves from an island to Aozora City, to live with her mom. There, she meets Laura, a mermaid, who makes her a Pretty Cure using the power of makeup. Manatsu and her fellow Pretty Cures fight against the servants of the Witch of Delays, who stole the motivation power from Laura's Mermaid Kingdom and are now trying to steal it from the human world.

The secret world "Healing Garden" has been healing the Earth. One day, Villains called "Byogens" attack Healing Garden with the goal to destroy Earth, which is now in big trouble! In order to save Earth, three "Earth doctor trainee" Healing Animals escape from Healing Garden to look for their partners. Along with them is Latte, the princess of Healing Garden, who holds a special power. When three ordinary girls meet the Healing Animals, they are transformed into Pretty Cures in order to face and defeat the Byogens.

Second year junior high school girl Hana Nono wants to be a cool big sister. Her first day at her new school gets off to a rocky start, but she meets some nice people and is looking forward to the next day when a mysterious baby named "Hug-tan" falls out of the sky along with her hamster caregiver Hari. Hug-tan is pursued by the evil Clias Company and its monsters that want her Mirai Crystal. Soon, Hug-tan is attacked but Hana's desire to protect the baby creates a new Mirai Crystal and she transforms into Cure Yell. Hana learns that the Mirai Crystals have the power to create tomorrow and if the crystals are taken, everyone's future will disappear.

Earth is once again under attack! Moebius, a dark lord from a parallel world called Labyrinth, is on a mission to conquer all of the worlds, and only the legendary Pretty Cure warriors can stop him. Now, between their studies and rehearsing to become idols, the trio must help put a stop to Moebius’s plans!

Nozomi Yumehara, a regular student, finds a magical book called the Dream Collet in the library and meets Coco and Nuts, two creatures from the Palmier Kingdom. They plead with Nozomi to restore their world, which has been destroyed by an organization called the Nightmares, by completing the Dream Collet and finding the 55 Pinkies to make any wish come true. Meanwhile, the Nightmares are moving into the real world. Once Nozomi agrees to help, Coco and Nuts transform her into the magical girl Cure Dream and turn four fellow students into her Pretty Cure team.

Somewhere in the universe there exists a place called Märchenland, where characters from fairy tales mingled together. Near the corner of Märchenland exists a world called Bad End Kingdom, where the antagonists of all fairy tales gathered. However, the Evil Emperor Pierrot of the Bad End Kingdom decided to invade the country and sealed away its Queen, while his minions traveled to Earth to collect Bad Energy and give the whole world the "Worst Ending". In order for the Queen to be revived and prevent the "Worst Ending" from happening, Candy, a fairy from Märchenland must gather the 5 legendary warriors called Pretty Cures to collect the Cure Décor, tokens of happiness that has been stolen and hidden away during the kingdom's attack. To gather them, Candy follows the 5 Beams of Light to Earth and it's there that she meets Miyuki Hoshizora, a transfer student from Nanairogaoka Middle School, who is already running late on her first day of class...

The Trump Kingdom is a magical world where everyone's hearts live happily with the guidance of their ruler Princess Marie Ange. However one day, a evil force known as the Jikochuu attacked the Kingdom. But a warrior who served Marie Ange, known as Cure Sword, fended the Jikochuu by herself before being sent the human world with her fairy partner to find the missing princess. The Cure Sword also brought with her three other fairies who are to find three chosen girls to be her allies. One of the fairies, Charles, finds a girl named Mana Aida who is her school's student council president. Whilst visiting the Clover Tower's observatory during her school's orientation day, Mana encounters a Jikochuu who attempts to manipulate her heart but being found by Charles as he gives her the power to become Cure Heart. Joined by Cure Diamond, Cure Rosetta and Cure Sword, the Dokidoki PreCure must fight to protect everyone's hearts from the Jikochuu.

The anime depicts the excitement of dorm life, coming of age while living with friends, and the anticipation and anxiety of a new life among roommates, separated from family. One day, the "Princess Precures" were revived by the Princess Perfume (and Kanata's dress-shaped "Dress-Up Key" that unlocks the Princess Perfume's power), and Haruka transforms into the flower princess Cure Flora. When unleashing their special signature moves, the Princess Precures' outfits turn into magnificent princess-like Mode Elegant long dresses.
